Others | Semaphorins are a family of phylogenetically conserved soluble and transmembrane proteins. Semaphorins were originally identified as axon guidance factors during neuronal development. Semaphorins are now known to be widely expressed mediators that play significant roles in immune responses and organ morphogenesis. Sema7A (also known as CD108) is a glycosylphosphatidylinositol-anchored semaphorin homologous to AHVsema. Sema7A was originally discovered as the John-Milton-Hagen human blood group antigen. Sema7A specially binds to VESPR/CD232/plexin-C1, which confirm that semaphorin is cellular counterpart of AHVsema. Sema7A transcripts are detectable in the embryonic nervous system and in adult tissues, including the brain, spinal cord, lung, and secondary lymphoid organs. Among lymphocyte populations, Sema7A is expressed mainly in activated T cells. Sema7A promotes axon outgrowth through β1-integrin receptors and contributes to the formation of the lateral olfactory tract. Sema7A expressed on activated T cells stimulates macrophages to produce proinflammatory cytokines through the α1β1 integrin. Sema7A-deficient mice are defective in T cell-mediated immune responses such as contact hypersensitivity and EAE. |
